HPLC法分析八种蜂蜜中的有机酸含量


本研究的目的在于用HPLC建立一种高效准确分析8种蜂蜜中10种有机酸的方法。通过采用Agilent 5 TC-C18(2)250×4.6mm色谱柱,3%甲醇和浓度为0.01mol/L的磷酸二氢钾溶液为流动相(体积比为3:97),磷酸调节PH为2.8,1.0mL/min的流速,10μL的进样量,柱温取室温,紫外检测波长为210nm时,各有机酸能够很好的分离。结果表明:蜂蜜中葡萄糖酸的含量最高(9.5746mg/mL),乙酸次之(1.1818mg/mL),马来酸的含量最低(0.0067mg/mL)。各种蜂蜜中有机酸总含量最高的是龙眼蜜(2.4169mg/mL),其次是椴树蜜(2.3395mg/mL)、荔枝蜜(2.1378mg/mL)、党参蜜(1.9772mg/mL)、紫云英蜜(1.9513mg/mL)、洋槐蜜(1.8768mg/mL)、荆条蜜(1.5382mg/mL)、桂花蜜(1.3187mg/mL)。含有机酸种类最多的蜂蜜是紫云英蜜和荔枝蜜,其次是椴树蜜、洋槐蜜、龙眼蜜、荆条蜜,种类最少的是党参蜜和桂花蜜,8种蜂蜜中都含有葡萄糖酸,甲酸也是蜂蜜中重要的有机酸,只有椴树蜜中没有检出。该种方法相对标准偏差小,在0.13%~2.30%范围内;回收率较高,在92.9%~109.8%范围内;各有机酸的R值(线性相关系数)较高,大于0.9985,线性范围在0.004~3.522mg/mL范围内。最后得出该方法灵敏度、准确度较高,重现性好,速度快,操作简单自动化,完全适用于蜂蜜中有机酸的测定分析。

Analysis of organic acids in eight kinds of honey by HPLC
 

Abstract: The purpose of this study is to establish an efficient and accurate method for the determination of 10 organic acids in eight honey by HPLC. Contains the most organic acids are honey and lychee honey, followed by linden tree honey, acacia honey, longan honey, Vitex, the least species is Codonopsis and sweet-scented osmanthus honey, 8 kinds of honey contains gluconic acid, Formic acid is also an important organic acid in honey, only linden tree honey is not detected. The relative standard deviation of this method is in the range of 0.13% ~ 2.30%, and the recovery is higher in the range of 92.9% ~ 109.8%. The R value (linear correlation coefficient) of each organic acid is higher than 0.9985, the linear range In the range of 0.004 to 3.522 mg / ml. Finally, the method has the advantages of high sensitivity, high accuracy, good reproducibility, fast speed and simple operation, and is suitable for the determination of organic acids in honey.
