拮抗放线菌对腐霉菌的生防作用筛选及菌株鉴定

腐霉菌(Pythium)是一种对植物有严重危害的植物病原菌,侵染极快,危害极强。长期以来,腐霉菌的防治主要是使用化学药剂,但已有报道常规杀菌剂甲霜灵的使用效果已有明显下降。本文利用从土壤中分离出的放线菌对腐霉菌进行平板对峙实验,确定放线菌LXD-1对腐霉菌有良好的拮抗作用。设计引物进行PCR扩增获得其16S rDNA, 将PCR产物纯化后进行测序分析, 其16S rDNA长度为1640bp。结合NCBI网站在线进行序列比对及系统发育进化树构建确定该放线菌的分类学地位,确定该放线菌为链霉菌(Steptomyces)。本研究对研究腐霉菌的生物防治提供基础,对生物农药的开发提供一定的理论支持。

Screening and Identification of Antagonistic Actinomycetes Against Pythium
 
Abstract:Pythium can cause serious plant disease, which infects rapidly, and causes a extensive damage. It has a long time to prevent Pythium by using chemical agent. However, it was reported that, the effect of fungicide such as metalaxyl has declined significantly. In this research, we used the actinomycetes isolated from soil to carry out antagonistic culture. The results showed that LXD-1 has a strong inhibition effect against Pythium. Primers were designed to amplify 16S rDNA. Then the PCR product was purified and sequenced. And 1640bp of 16S rDNA sequence was obtained. According to the results of alignment by NCBI online software and the phylogenetic tree, the strain LXD-1 was identified as Steptomyces. The results will provide base for biological control of Pythium, and provide support for the development of biological pesticide.
