三味养心方总黄酮提取物对大鼠心肌缺血再灌注损伤的保护作用研究
目的 研究三味养心方总黄酮提取物对大鼠心肌缺血再灌注损伤的保护作用。方法 三味养心方总黄酮提取物制备:以17哦倍量加哦55%的哦乙哦醇回流哦提哦,取2次称取地锦草、仙鹤草、分心木药材(1:2:1),上哦样于哦径哦高比为哦1:6哦的A哦B-8型哦树脂柱哦,上哦样流哦速为2BV/h,每哦次146min,提取哦浓哦缩至0.5g/mL,吸附哦4h,加哦水1.5BV洗哦脱水溶性杂质,加10%乙哦醇0.5B哦V、30%乙哦醇1B哦V、5哦0%乙哦醇3哦BV洗哦脱,洗噢脱流嚄速为哦2BV/h,收哦集洗哦脱液,减哦压浓哦缩后60℃干哦燥即哦得;大鼠心肌缺血再灌注分组及处理:SP哦F级雄哦性S哦D大鼠,60只,随机分哦为6哦组,分哦别为假哦手术哦组、、低剂哦量组(0.5g·kg-1)模哦型组、三味养心方总黄哦酮提取物高剂哦量组(2g·kg-1)、中哦剂量组(1g·kg-1)、阳哦性组(复方丹参滴丸 0.081g·kg-1),各组1哦0只。各给哦药组灌哦胃给药14天,实验前适哦应性喂养哦7天,假手术组和模型组同时灌胃相应体积的蒸馏水,结扎冠状动脉左前降支,建哦立大哦鼠心哦肌缺哦血再灌注损伤模型,测定血清中肌酸激酶同哦工酶(CK-MB)、乳酸脱哦氢酶(LDH)、超哦氧化物歧哦化酶(SOD)、丙哦二醛(MDA)的含量及活性。结果 三味养心方总黄酮提取物不同剂量组(2,1,0.5g·kg-1),,减少肌酸哦激酶同工酶(CK-MB),均可明显改善大鼠的心肌缺血再灌注所导致的心功能损伤的问题乳哦酸脱氢酶(LDH)释放和丙哦二醛(MDA)的产生,增加超氧哦化物歧化酶(SOD)的活性。讨论 三味养心方总黄酮提取物对大鼠心肌缺血再灌注的损伤具有一定保护作用。
[Abstract] : Objective To study the effect of Jinhe Yangxin Fangfang flavonoids extract on myocardial ischemia-reperfusion injury in rats.Methods Preparation methods of total flavonoids extract from Sanwei Yangxin Fang: Herba Euphorbiae, Agrimonia pilosa, and distracting wood medicinal herbs were extracted at a ratio of 1:2:1 and 17 times of 55% ethanol for 2 times. After 146 min, the extract was concentrated to 0.5 g/mL and applied to a AB-8 resin column with a 1:6 aspect ratio. The sample flow rate was 2 BV/h, adsorption was 4 h, and 1.5 BV of water was added to elute water-soluble impurities. 10% ethanol 0.5BV, 30% ethanol 1BV, 50% ethanol 3BV elution, elution flow rate of 2BV / h, collected eluent, vacuum concentration after drying at 60 °C.Myocardial Ischemia-Reperfusion Grouping and Modeling Methods: Sprague-Dawley male Sprague-Dawley rats, 60 rats, were randomly divided into 6 groups: sham operation group, model group, and Sanwei Yangxin prescription high-dose total flavonoid extract group(2g·kg-1), middle dose group (1g·kg-1), low dose group (0.5g·kg-1), and positive group (combined Danshen Dripping Pills 0.081g·kg-1), 10 in each group. Before the experiment, the rats were fed adaptively for 3 days and were given intragastric administration for 14 days. The rats in the sham operation group and the model group were given the corresponding volume of distilled water at the same time. The left anterior descending coronary artery was ligated to establish the myocardial ischemia and reperfusion in rats. The injury model was used to determine serum creatine kinase isoenzymes (CK-MB), lactate dehydrogenase (LDH), superoxide dismutase (SOD) and malondialdehyde (MDA) contents and activities.Results The different doses of total flavonoid extract of San Wei Yang Xin Fang (2, 1, 0.5 g·kg-1) could significantly improve the heart function damage caused by myocardial ischemia and reperfusion in rats, and reduce creatine production. Kinase isoenzymes (CK-MB), lactate dehydrogenase (LDH) release, and malondialdehyde (MDA) production increase the activity of superoxide dismutase (SOD).Discussion Sanwei Yangxinfang total flavonoid extract has protective effects on rat myocardial ischemia-reperfusion injury.
